To determine the clinical chemical blood parameters, plasma enzymatic activities, and specific substrate and electrolyte levels, biochemical tests are performed with mouse blood.

These procedures are applicable to the mouse phenotyping platform at the RIKEN BRC. The tests should be performed by trained staff members who understand the safety and hygiene rules required by RIKEN. Any queries, comments, or suggestions, relating either to this SOP in general or to a specific problem encountered during a procedure, should be addressed to the Blood Tests Unit Leader. Any deviation from this protocol must be reported to the Blood Tests Unit Leader.

Testing is carried out in accordance with common animal handling procedures. Coveralls (in the rearing facility) or laboratory coats (in the laboratory facility) must be worn with gloves at all times in the work area. If animals are dropped on the floor, treat them in accordance with the safety and hygiene guidelines of the rearing facility at RIKEN. If something unexpected happens during the primary screening, it must be quickly reported to the Blood Tests Unit Leader and recorded.

Use commercial measurement kits for mouse samples, if available. Kits for human samples (clinical tests) may be used if mouse-specific kits are not commercially available. Make inquiries to the manufacturers because some clinical test reagents for enzymes, low-molecular-weight compounds, sugars, and lipids can be used to analyze mouse samples. Because they were not specifically made for measurements of mouse samples, however, their accuracy should be checked before using them for experimental analyses.

Before the analysis, an inspection of the automatic analyzer should be performed (see the manufacturer¥apos;s manual). Before the analysis, all parameters are tested with control serum [see Appendix (3) v2]. Values must lie within the quality control ranges, or within the ranges indicated on the documents describing the control sera. Maintenance of the analyzer is performed according to the manufacturer¥apos;s manual.

Prepare the samples according to the SOP for Blood Collection for Clinical Biochemical Analysis Use the manual for the automated serum analyzer (JCA-BM6070) and documents describing the test reagents for the analyses. After the analysis, data is transferred to the MUSDB database using the online network.
